Thermotolerant Zymomonas mobilis: Comparison of Ethanol Fermenta- tion Capability with that of an Efficient Type Strain
نویسندگان
چکیده
Zymomonas mobilis is an alternative microorganism to Saccharomyces cerevisiae for ethanol production. To find a thermotolerant Z. mobilis strain, the growth and ethanol production of four isolates in Thailand were compared with those of the efficient strain ZM4 (NRRL B-14023) at different temperatures. One of the selected strains, TISTR 405, was found to grow and produce ethanol even at 39 ̊C to an extent similar to that at 30 ̊C, and the growth and ethanol productivity at 39 ̊C were better than those of ZM4 at 30 ̊C, suggesting that TISTR 405 is suitable for ethanol fermentation at high temperatures. Analysis of genes directly related to ethanol formation or degradation, adhA, adhB and pdc, encoding alcohol dehydrogenase (Adh) A, AdhB and pyruvate decarboxylase, respectively, revealed that these genes were highly conserved in both strains. Comparison of their gene expression and activity of the products in both TISTR 405 and ZM4 at different temperatures or growth phases indicated that there was not a great difference at the transcriptional level, but the total activity of AdhA and AdhB in TISTR 405 was higher than that in ZM4. Both strains showed a significant increase in AdhB activity in the stationary phase.

The ice edge of the Barents Sea east of Svalbard is an area where the warm, salty water of the North Atlantic (AtW) interacts with cold, less dense, saline Arctic water (ArW) and the water produced by melting ice (MIW). Many of the CTD profiles (CTD stands for Conductivity-Temperature-Depth) obtained in this region by Norwegian Polar Institute expeditions in 1999 and 2007 contain layers that ar...
متن کاملEvaluation of Ethanol Production from Tannin-Reduced Carob Pod Extracts by Zymamonas Mobilis
Regarding some reported antimicrobial properties of tannins; Zymomonas mobilis was used to obtain ethanol from tannin-reduced carob pod extract (TR-CPE). Culture of 50 mL volume containing 7.5 g sugar at pH 5.5 and 0.03 g bacterial inoculums with shaking at 80 rpm was used. Using response surface methodology (RSM), the maximum ethanol concentration of 5.34 % w/v (higher than that reported earli...
متن کاملCellulosic Ethanol Production by Recombinant Cellulolytic Bacteria Harbouring pdc and adh II Genes of Zymomonas mobilis
The ethanol fermenting genes such as pyruvate decarboxylase (pdc) and alcohol dehydrogenase II (adh II) were cloned from Zymomonas mobilis and transformed into three different cellulolytic bacteria, namely Enterobacter cloacae JV, Proteus mirabilis JV and Erwinia chrysanthemi and their cellulosic ethanol production capability was studied. Recombinant E. cloacae JV was found to produce 4.5% and ...

Zymomonas mobilis ATCC 10988 is the type strain of the Z. mobilis subsp. mobilis taxon, members of which are some of the most rigorous ethanol-producing bacteria. Isolated from Agave cactus fermentations in Mexico, ATCC 10988 is one of the first Z. mobilis strains to be described and studied. Its robustness in sucrose-substrate fermentations, physiological characteristics, large number of plasm...
متن کاملThermotolerant genes essential for survival at a critical high temperature in thermotolerant ethanologenic Zymomonas mobilis TISTR 548
BACKGROUND High-temperature fermentation (HTF) technology is expected to reduce the cost of bioconversion of biomass to fuels or chemicals. For stable HTF, the development of a thermotolerant microbe is indispensable. Elucidation of the molecular mechanism of thermotolerance would enable the thermal stability of microbes to be improved. RESULTS Thermotolerant genes that are essential for surv...
